摘要
目的 探讨葡萄糖转运体SLC2 A1~9在K562细胞、正常人与白血病患者血细胞中的表达差异,分析其在白血病发生发展中的作用.方法 通过培养收集人慢性髓原白血病细胞(K562细胞)、正常人和白血病患者血细胞,采用反转录PCR和电泳技术检测SLC2A1~9 mRNA的表达量.结果 K562细胞可见SLC2A1、3、4、6、8、9 mRNA表达,正常人血细胞可见SLC2A3、6、8 mRNA表达,白血病患者血细胞可见SLC2A3、6、8、9 mRNA表达;白血病患者组SLC2A6、9 mRNA的表达水平均明显高于正常人组(P<0.05),而SLC2A3 mRNA的表达水平则明显低于正常人组(P<0.01),SLC2A8 mRNA的表达则两组间无显著差异(P>0.05).结论 SLC2A6、9在白血病中高表达,可能成为白血病基因诊断的分子标志物和抗癌治疗新靶点.
Objective To investigate the differential expression of glucose transporter SLC2A1 ~ 9 in K562 cells,normal blood cells and blood cells of leukemia patients,and analyze role of glucose transporters in the development and progression of leukemia.Methods The mRNA of SLC2A1~9 of human chronic myelogenous leukemia K562 cells,blood cells from healthy volunteers and patients with leukemia was detected with PCR.Results SLC2A1,3,4,6,8,9 mRNA expressed in K562 cells,SLC2A3,6,8 mRNA expressed in normal human blood cells and SLC2A3,6,8,9 mRNA expressedin leukemia patients blood cells.SLC2A6,9 mRNA expression levels of leukemia patients were significantly higher than that of the normal group (P<0.05),whereas the expression of SLC2A3 mRNA levels were significantly lower than that of the normal group (P<0.01).There is no significant difference of SLC2A8 mRNA expression between the two groups (P>0.05).Conclusion The SLC2A6,9 are high expressed in leukemia and their expression may be the molecular markers of leukemia and new targets for anti-cancer therapy.
