胰岛素样生长因子-Ⅰ、肝细胞生长因子、转化生长因子-β1、干扰素-γ对奶牛乳腺上皮细胞增殖活性的影响 被引量:6

摘要 本试验应用组织块培养和差速消化法获取原代奶牛乳腺上皮细胞,免疫荧光鉴定正确后MTT法评价不同浓度的胰岛素样生长因子-I(insulin-like growth factor-I,IGF-I)、肝细胞生长因子(hepatocyte growth factor,HGF)、转化生长因子-β1(transforming growth factor-β1,TGF—β1)、干扰素-y(interferon-y,IFN-y)对奶牛乳腺上皮细胞体外增殖的影响。结果表明,IGF-I、HGF分别在10~200和0.1~100ng/mL对乳腺上皮细胞的增殖呈正相关,而TGF-β1(2.5~100ng/mL)、IFN-y(5~160ng/mL)则剂量依赖性地抑制乳腺上皮细胞增殖。提示,IGF—I、HGF均能促进奶牛乳腺上皮细胞的体外增殖,该作用在一定浓度范围内有剂量依赖性;TGF-β1、IFN-y对乳腺上皮细胞的增殖作用出现剂量抑制效应。 This experiment was conducted to investigate the regulation of insulin-like growth factor- I (IGF- I ), hepatocyte growth factor (HGF), transforming growth factor-β1 (TGF-β1) and interferon-y (IFN-y) in the growth of bovine mammary epithelial cells in vitro. The bovine mammary epithelial cells were isolated by explant culture technique and differential adhesion method and identified by immunoehemistry. The effect of cytokines regulating the growth of bovine mammary epithelial ceils in different concentrations was measured by MTT. The results showed that when the concentrations of IGF- I and HGF were 10 to 200 and 0. 1 to 100 ng/mL, respectively, the growth of bovine mammary epithelial cells increased. While when the concen trations of TGF-β1 and IFN-y were 2.5 to 100 and 5 to 160 ng/mL, respectively, the growth of bovine mammary epithelial cells decreased in a dose-dependent manner. These results indicated that IGF I and HGF could upregulate the proliferation of bovine mammary epithelial cells in vitro in a dose dependent manner. TGF-β1 and IFN y could inhibit the proliferation of bovine mammary epithelial cells in vitro.
