摘要
目的:探讨姜黄素诱导乳腺癌细胞MDA-MB-231凋亡的可能机制。方法:分别采用0,25,50,100,150,200μmol·L-1浓度的姜黄素对乳腺癌细胞MDA-MB-231进行干预,采用MTT法观察其对乳腺癌细胞增殖的影响,采用流式细胞术观察其对乳腺癌细胞凋亡的影响,采用实时定量PCR及Western blot检测乳腺癌细胞内葡萄糖调节蛋白78(GRP78)及C/EBP同源蛋白(CHOP)表达水平。结果:姜黄素可通过诱导乳腺癌细胞MDA-MB-231抑制其增殖能力,并呈现浓度依赖性;姜黄素可显著升高乳腺癌细胞内GRP78及CHOP表达水平。结论:姜黄素可通过内质网应激途径诱导乳腺癌细胞MDA-MB-231的凋亡。
Objective: To explore the possible mechanism of curcumin in inducing the apoptosis of breast cancer cell MDA- MB-231. Method: Curcumin of different concentrations at 0,10 25,50,100,150,200μmol L-1 were used to intervene breast cancer cells MDA-MB-231 for 24 hours. MTT was used to observe its effect on the proliferation of breast cancer cells. The flow cytometry was used to detect its effect on the cell apoptosis. The real-time quantitative PCR and Western blot was used to assess the expression levels of GRP78 and CHOP in breast cancer cells. Result: Cureumin could inhibit the proliferative ability of breast cancer cells by inducing them in a concentration-dependent manner. Curcumin could significantly increase the expression levels of GRP78 and CHOP in breast cancer cells. Conclusion: Curcumin could induce the apoptosis of breast cancer cells MDA-MB-231 by activating endoplasmic reticulum stress.
