抵当汤对阿尔茨海默病大鼠脑组织端粒酶活性的影响 被引量:4

The influence of Didang decoction on telomerase activity in brain tissue of rats with Alzheimer disease

摘要 目的观察抵当汤对阿尔茨海默病(AD)大鼠脑组织端粒酶活性及学习记忆能力的影响。方法 40只雄性Wistar大鼠随机分为假手术组、模型组、多奈哌齐组和抵当汤组。采用右侧海马注射Aβ25-35方法制备AD大鼠模型,治疗组采用抵当汤治疗,对照组给予多奈哌齐,均连续灌胃给药,疗程为8周,采用跳台实验观察各组大鼠学习记忆能力的改变。实验结束后处死大鼠,采用ELISA检测脑组织端粒酶活性。结果实验结束后各用药组逃避潜伏期较模型组明显延长,错误次数明显减少(P<0.05,P<0.01),抵当汤组较多奈哌齐组错误次数减少更明显(P<0.05);与模型组比较,抵当汤组和多奈哌齐组脑组织端粒酶活性明显提高(P<0.05,P<0.01),抵当汤组较多奈哌齐组提高更明显(P<0.05)。结论抵当汤可显著改善AD大鼠的学习记忆能力,其机制可能与该方提高大鼠脑组织端粒酶活性有关。 Objective To observe the influence of Didang Decoction on telomerase activity and the ability of learning and memory of brain tissue in rats with Alzheimer disease (AD). Methods 40 male Wistar rats were randomly divided into the pseudo operation group,model group,donepezil group and Didang decoction group.AD rats model was made by the method of injecting A beta 25-35 in the right hippocampus.The Didang decoction group was treated by used Di- dang decoction, the donepezil group was treated by donepezil,the rats,all groups were given continuous intragastric ad- ministration for 8 weeks,after the medication step down test was performed to observe the changes of learning and memory ability in different groups of rats.After the experiment,the rats were sacrificed and ELISA was used to detect telomerase activity in brain tissue. Results Compared with the model group,the escape latent period of rats in each treatment group was prolonged significantly (P〈0.05,P〈0.01),error number was decreased obviously,it was more signifi- cantly decreased in the Didang decoction group compared with that in the donepezil group (P〈0.05);compared with the model group,the telomerase activity of the Didang decoction group and donepezil group increased significantly (P〈0.05, P〈0.01),and it was more significantly increased in Didang decoction group compared with that in the donepezil group (P〈0.05). Conclusion Didang decoction can significantly improve the learning and memory ability of rats with AD,and its mechanism may be related to this prescription increasing telomerase activity in brain tissue of rats.
