摘要
目的獉獉:利用化学方法将丁丙诺啡半抗原与载体蛋白偶联制备丁丙诺啡人工抗原。方法獉獉:结构修饰后的丁丙诺啡半抗原与载体BSA蛋白经过戊二醛法联接合成丁丙诺啡人工抗原,利用紫外光谱扫描法、蛋白质电泳法和胶体金免疫层析法进行检测。结果獉獉:丁丙诺啡人工合成抗原联接成功,胶体金免疫层析检测结果显示,合成抗原与抗体有特异性反应,具有活性,交叉反应良好。结论獉獉:该方法合成的丁丙诺啡抗原可作为丁丙诺啡免疫用抗原和丁丙诺啡免疫检测方法建立所用包被抗原。
Objective.: To couple the hapten -buprenorphine with carrier protein to prepare buprenorphine artificial antigen. Methods: Modified buprenorphine was coupled with carrier bovine serum albumin (BSA) by glutaraldehyde crosslinking method to produce the artificial antigen. Subsequently, this artificial antigen was detected by UV scan, electrophoresis and gold immunochromatography assay. Results: Buprenorphine artificial antigen was successfully synthesized and produced specific reaction with antibody and demonstrated a good cross - reactivity by gold immunochromatography assay. Conclusion: The prepared buprenorphine artificial antigen can be used as immunogen and as the coating antigen in immunological assay.
