摘要
目的观察IgA肾病大鼠模型血清白介素-2(IL-2)和血清白介素-6(IL-6)变化对肾纤维化的影响。方法根据IgA肾病大鼠模型的血清IL-2和IL-6浓度各设置3个浓度梯度(IL-2:10、30和90 pg/mL;IL-6:100、300和600 pg/mL),分别单独和联合干预肾小管上皮细胞(RTEC),采用免疫组织化学法检查转化生长因子β1(TGF-β1)在各组细胞的表达。结果 IL-2单独干预对肾小管上皮细胞TGF-β1表达无影响(P<0.05);而IL-6能直接增强肾小管上皮细胞TGF-β1表达(P<0.01)。IL-2和IL-6对肾小管上皮细胞TGF-β1表达存在交互作用;当IL-2=90 pg/mL,IL-6=300 pg/mL时,TGF-β1表达量最高(P<0.01)。结论血清IL-2和IL-6变化,特别是IL-6的升高可能与TGF-β1介导的IgA肾病肾纤维化有关。
[Objective] To evaluate the effects of interleukin- 2 (IL- 2) and interleukin- 6 (IL- 6) changes in IgA nephropathy rats model on renal fibrosis. [Methods] Renal tubular epithelial cells (RTEC) were divided into different groups and treated with IL- 2 and IL- 6 by different concentration (IL- 2: 10, 30, 90 pg/mL; 1L- 6: 100, 300, 600 pg/mL) individually and mixed together, the expression of transforming growth factor beta 1 (TGF- B,) in each group was detected with immunohistochemical methods. [Results] IL -2 had no effect on the expression of TCF- B1 in RTEC individually (P 〉0.05). However, IL- 6 could affect RTEC TGF-B1 expression directly (P〈0.01). IL-2 combined with IL-6 had interaction on rat kidney TGF- B1 expression. When the concentration of IL-2 was 90 pg/mL together with IL - 6 was 300 pg/mL, TGF - B1 had highest expression levels (P〈0.01). [Conclusion] Serum IL-2 and IL-6 changes, especially IL-6 increased, may promote the development of lgA nephropathy TGF- B1- mediated renal fibrosis.
