摘要
目的探讨HSP70参与体温调控的作用及其可能机制。方法复制家兔LPS发热模型,RT-PCR和Western blot检测在发热过程中单核细胞IL-1β、TNF-αmRNA与HSP 70的表达。结果注射LPS0.5μg/kg后家兔体温明显升高,出现两个体温高峰;由LPS引起的发热过程中单核细胞TNF-α、IL-1βmRNA的表达水平分别在60 min和180 min达高峰,400 min以内降至基础水平;单核细胞HSP70的含量在注射LPS后160 min时开始升高,实验结束时达最高值;LPS引起发热的过程中单核细胞HSP70含量与单核细胞TNF-α、IL-1βmRNA的表达量之间呈负相关关系。结论 LPS致发热时,HSP70可能通过抑制TNF-α、IL-1β等内生致热原基因的表达而限制体温升高。
[Objectives] To investigate the effect of heat shock protein 70 and its mechanism in thermoregulation. [Methods] The relationship among the expression of HSP70 and interleukinl β (IL-lβ) mRNA and tumor necrosis factor et (TNF-α) mRNA in mnnocytes was studied during the different fever stages in rabbit fever model induced by LPS. The expression of IL-1β and TNF-ot mRNA were measured by RT-PCR, HSP 70 expression was measured by Western blot. [Results] Intravenous injection of LPS produced a double peak temperature arisen at 60 min and 180 rain. The expression of 1L-lβ mRNA in monocytes had a peak at 160 rain, while the peak of TNF-α mRNA expression occurred at 80 rain after intravenous injection of LPS. The content of the HSP70 increased after 160 rain intravenous injection of LPS, reached the peak value at 400 min. The results indicated that the content of the liSP70 was negative correlation with the expression of IL-1β mRNA and TNF-α mRNA. [ Conclusions ] It is possible thai HSP70 limits the rise in body temperature by repressing the gene expressions of the endogenous pyrogen, IL-1 and TNF-α.
