阿司匹林诱导人宫颈癌Siha细胞凋亡及其抑制作用的研究 被引量:1

摘要 目的研究阿司匹林(aspirin)对宫颈癌Siha细胞凋亡和增殖的机制。方法培养人宫颈癌Siha细胞,加入不同浓度阿司匹林(1.0 mmol/L、5.0 mmol/L、10.0 mmol/L)干预培养,HE染色观察Transwell小室穿梭能力;划痕愈合实验观察迁移能力;应用流式细胞技术检测Siha细胞凋亡和细胞周期的变化;应用Giemsa染色观察细胞形态学的改变;TUNEL染色检测细胞凋亡情况;蛋白质印迹法检测阿司匹林对宫颈癌Siha细胞bcl-2及bax蛋白表达的影响。结果阿司匹林表现出明显地抑制人宫颈癌Siha细胞的迁移、增殖以及促进凋亡(P<0.05),且伴随浓度的递增和时间的延长,抑制作用更明显。结论阿司匹林具有较强的抗人宫颈癌作用,其机制可能与阿司匹林诱导宫颈癌细胞凋亡,并抑制其增殖有关。 [ Objective ] To assess the effects of aspirin on the proliferation and apoptosis of human cervical can- cer Siha cell line. [Methods ] Siha cell was cultured and induced with concentration gradientof aspirin.HE staining was used to observe transwell chamber shuttle capacity, scratch healingexperimental for migration. The effect of as- pirin on Siha cell apoptosis and cell cycle was observed by cell flow eytometry (FCM). The structures of cell were observed under light microscope with Giemsa stain, and the apoptosis was shown wilh TUNEL staining. Expression of bel-2 and bax protein was determined by Western blotting. [ Results ] The results showed aspirin significantly in- hibit human cervical cancer Siha cell migration, proliferation and promote apoptosis (P 〈0.05), and with increasing concentration and time extension, the trend is more obvious. [ Conclusions ] Aspirin has strong inhibition on cervical cancer, its mechanism may be related to induction of apoptosis on cervical cancer, and inhibition of proliferation.
