摘要
目的观察丹参酮ⅡA磺酸钠(sodium tanshinone ⅡA sulfonate,STS)对转换生长因子-β1(transforminggrowth factor-β1,TGF—B1)诱导的心房成纤维细胞分化的影响及其机制。方法体外培养1~2d龄乳鼠心房成纤维细胞,酶联免疫吸附测定法检测培养上清液中Ⅰ/Ⅲ型胶原含量,四甲基偶氮唑蓝法检测细胞活力,免疫荧光法及蛋白质印迹法测定α-平滑肌肌动蛋白表达,同时蛋白质印迹法检测细胞外调节蛋白激酶1/2磷酸化。结果转化生长因子-β1能显著提高心房成纤维细胞培养上清液中Ⅰ/Ⅲ型胶原含量,提高细胞增殖活力,同时上调α-平滑肌肌动蛋白表达及细胞外调节蛋白激酶1/2磷酸化。以上各指标能被丹参酮ⅡA磺酸钠(3,10,30μmol.L^-1)逆转。结论丹参酮ⅡA磺酸钠可以抑制转化生长因子-β1诱导的心房成纤维细胞分化,机制可能与下调细胞外调节蛋白激酶1/2活化有关。
OBJECTIVE To observe the effects and mechanisms of sodium tanshinone Ⅱ A sulfonate (STS) on transforming growth factor-β1 (TGF-β1)-induced atrial fibroblasts differentiation. METHODS In the culture of neonatal rat atrial fibro-blasts, type Ⅰ/Ⅲ collagen in the cell culture supernatant were measured by enzyme-linked immunosorbent assay. The prolifera-tion activity was measured by methyhhiazolyl tetrazolium(MTT) assay. Atrial fibroblasts differentiation was determined by meas-uring the expression of α-SMA( α-smooth muscle actin) using Western blot and immunofluorescence. Extracellular signal-regula-ted kinase 1/2(ERK1/2) activation was measured by Western blot. RESULTS STS can decrease TGF-β1-induced elevations of type Ⅰ/Ⅲ collagen and proliferation activity; inhibit the expression of α-SMA and ERK1/2 activation. CONCLUSION STS can inhibit TGF-β1-induced atrial fibroblasts differentiation, the mechanism may be associated with depression of ERK1/2 signaling pathway.
