HPLC测定八正胶囊中的大黄素与大黄酚

Determination of emodin and chrysophanol in Bazheng capsules by HPLC

目的用HPLC法测定八正胶囊中的大黄素、大黄酚。方法采用菲罗门C18色谱柱（250mm×4．6mm，5μm），流动相为乙腈-甲醇-0．1％磷酸溶液（42：23：35），流速1．0mL·min-1，检测波长254nm。结果大黄素、大黄酚的线性范围分别为0．0267～0．4272μg（r=0．9998）、0．0417～0．6677μg（r=0．9999），总大黄素、总大黄酚、游离大黄素、游离大黄酚的平均加样回收率分别为97．35％（RSD=1．7％）、98．39％（RSD=0．4％）、100．70％（RSD=1．20％）、104．41％（RSD=0．9％）。结论所用方法专属性好，准确、重复性好，可作为八正胶囊质量控制的方法之一。

OBJECTIVE To establish an HPLC method to determine the contents of emodin and chrysophanol in Bazheng capsules. METHODS Phenomenex - C18 column（250 mm × 4.6 mm,5 μm） was used. Acetonitrile - methanol - 0.1% phosphoric acid solu- tion（42 ： 23 ： 35） was used as the mobile phase at a flow rate of 1.0 m.rain-1, and the detection wavelength was 254 nm. RESULTS The calibration curves showed good linearity in the range of 0. 0267 -0. 4272μg （emodin）,0. 0417 -0. 6677 μg （ehrysophanol）. The average recoveries were 97.35 % （RSD = 1.7 % ） ,98.39% （RSD = 0.4% ） , 100.70% （RSD = 1.2% ） , 104.41% （RSD =0. 9% ） ,respectively. CONCLUSION The method is accurate and reproducible,which can be used in the quality control of Bazheng capsules.
