应用填充床式生物反应器制备猪圆环2型病毒原液

Application of Fix-bed Bioreactor for Production of Porcine Circovirus Type2 Bulk

[目的]应用填充床式生物反应器培养PK-15细胞制备PCV2病毒原液。[方法]以PK-15细胞为基质,PCV2 YZ株为毒种,利用5L生物反应器进行灌流培养,连续收获病毒液16 d。[结果]细胞密度最高到1.9×107个/ml,收获滴度106.83TCID50/ml的病毒原液62.5L。[结论]应用填充床式生物反应器可以制备高质量的PCV2病毒原液。

[ Objective] To prepare PCV2 bulk with PK-15 cells in fix-bed bioreactor. [ Method] PCV2 was cultured in PK-15 ceils in 5 L fix- bed bioreactor, continuously harvested for 16 days; [ Result] The cell density reached 1.9 × 107 cells/ml at most, and the viral titer of the to- tal of 62.5 L harvest could be as high as 106.83 TCID50/ml. [ Conclusion] Fix-bed bioreactor with Fibra-Cel carriers may be used for prepara- tion of PCV2 bulk with high quality.
