参芪益心方对阿霉素诱导的慢性心力衰竭大鼠心肌结构和细胞因子的影响 被引量:12

Effects of Shenqi Yixin Fang on Myocardial Structure and Cytokines in Doxorubicin-induced Chronic Heart Failure Rats

摘要 目的:观察参芪益心方对慢性心力衰竭(CHF)大鼠心肌形态、心功能、血浆中血管紧张素Ⅱ(AngⅡ)、肿瘤坏死因子-α(TNF-α)的影响。方法:将90只Wistar雄性大鼠随机平均分为6组:正常对照组、模型组、依那普利对照组、芪苈强心对照组、参芪益心方低剂量(16.96 g·kg-1·d-1)组、高剂量(33.92 g·kg-1·d-1)组。利用阿霉素第l,3,5周,3 mg·kg-1;第2,4,6周,2 mg·kg-1的剂量,每日1次,灌胃4周,观察大鼠的心肌组织形态学及超微结构变化,运用超声心动图检测大鼠左室收缩末期内径(LVSD)、左室舒张末期内径(LVDD)、左室射血分数(EF)、左室短轴缩短率(FS),放免法测定大鼠血浆AngⅡ,TNF-α。结果:经EF,FS提示造模成功,各治疗组LVSD,LVDD,EF,FS均有改善,参芪益心方高剂量组、中药对照组、西药对照组与模型组比较有统计学意义(P<0.05)。各治疗组AngⅡ,TNF-α均有不同程度的降低,与模型组比较有统计学意义(P<0.05)。结论:参芪益心方可以改善CHF大鼠心肌组织形态,抑制心室重构,改善CHF大鼠心功能。 Objective:To observe the effect of the Shenqi Yixin Fang on cardiac function,plasma angiotensin Ⅱ (Ang Ⅱ) and tumor necrosis factor-α (TNF-α) of chronic heart failure (CHF) rats.Method:Ninty male Wistar rats were randomly divided into six groups:normal control group,model group,enalapril control group,the Qili Qiangxin control group,the low dose of Shenqi Yixin Fang group (16.96 g ·kg-1·d-1),the high dose of Shenqi Yixin Fang group (33.92 g ·kg-1·d-1).Doxorubicin was used to induce CHF models,and the modelling rats were given drug intervention for 4 weeks.The echocardiography was used to measure cardiac function,plasma Ang Ⅱ and TNF-α were measured by radioimmunoassay.Result:In model group the left ventricular end-systolic diameter (LVSD),left ventricular end-diastolic diameter (LVDD),left ventricular ejection fraction (EF),left ventricular fractional were shorten compared with normal control group (P < 0.05).In treatment group,these indicators were improved compared with modek group (P < 0.05).In treatment groups the plasma Ang Ⅱ/TNF-α levels were reduced compared with the model group (P < 0.05).Conclusion:The Shenqi Yixin Fang can improve myocardial tissue morphology,inhibit ventricular remodeling and improve cardiac function in CHF rats.
