摘要
目的:探讨过敏性休克小鼠肺组织中胱硫醚-γ-裂解酶(CSE)的表达。方法:取健康昆明种小鼠36只,随机分为过敏性休克模型组和磷酸盐缓冲液(PBS)对照组,每组再分为死亡即时组、冷冻48 h组和冷冻7 d组,每组6只,模型组通过注射卵蛋白(OVA)制作过敏性休克模型,致敏第3周诱发过敏性休克致死或颈椎脱臼处死动物。对照组采用PBS代替OVA,第3周与模型组小鼠一并处死。各组小鼠分别在3个时间点取肺组织,福尔马林固定,制作石蜡切片,并行免疫组织化学染色检查观察CSE在肺组织中的表达情况。结果:模型组各时间点肺组织中见胱硫醚-γ-裂解酶主要表达在肺泡上皮细胞胞浆,对照组肺泡上皮细胞呈弱阳性。结论:过敏性休克小鼠肺组织中有胱硫醚-γ-裂解酶表达,死后冷冻7 d之内,肺组织中胱硫醚-γ-裂解酶表达随着时间的延长呈下降趋势,应用免疫组化染色检查肺组织中胱硫醚-γ-裂解酶的表达可为过敏性休克死亡提供一定的法医病理学诊断依据。
Objective:To study the expression of cystathionine-γ-lyase (CSE) in lung tissue of mice with anaphylactic shock. Methods :36 healthy Kunming mice were randomly divided into anaphylactic shock model and phosphate-buffered saline (PBS) control group, each group was further divided into immediate death, frozen 48 h and frozen 7 d group, with 6 mice in each group. The anaphylactic shock model was reprgduced by injection of ovalbumin (OVA) ,three weeks after sensitization, OVA was injected into the heart of the mice to induce anaphylactic shock and death, or killed by cervical dislocation. The control group with PBS instead of OVA, the mice were killed in the third week in conjunction with the model mice. The lung tissues were taken out, fixed by Formalin, and then paraffin sections were made. Then pathologic changes of the lungs were observed under microscope, immunohistochemical staining method was used to detect CSE ex- pression in lung tissue sections. Results:Cystathionine-γ-lyase is mainly expressed in the cytoplasm of alveolar epithelial cells in lung tissue of model group, and alveolar epithelial cells were weakly positive in control group at each time point. Conclusion : Cystathionine-γ-lyase were expressed in the lung tissues of anaphylactic shock mice, 7 d frozen after death, the expression of cystathionine - γ-lyase in lung tissues decreased with time goes on. The detection of the expression of cystathionine-γ-lyase by immunohistochemical staining method may provide an objective method for forensic pathology diagnosis of anaphylactic shock.
