鸡血藤对去卵巢豚鼠脂代谢影响的实验研究

Experimental Study on The Effects of Suberect Spatholobus Stem on Lipid Metabolism of Ovariectomized Guinea Pig

目的:建立去卵巢豚鼠脂代谢紊乱模型,探讨鸡血藤提取物对去卵巢豚鼠脂代谢的影响。方法:采用去卵巢方法建立豚鼠脂代谢紊乱模型。30只雌鼠,随机分5组,每组6只:①正常对照组;②模型组;③阳性药物对照组(戊酸雌二醇,80mg/kg·d);④低剂量组(鸡血藤提取物,40mg/kg·d);⑤高剂量组(鸡血藤提取物,80mg/kg·d)。术后1周内所有去除卵巢豚鼠均连续阴道涂片检查。术后1周建模成功后,阳性药物对照组用戊酸雌二醇水溶液灌胃(80 mg/kg·d),鸡血藤提取物低剂量、高剂量组分别按40、80mg/kg·d剂量灌胃,正常对照组、模型组每天等体积生理盐水灌胃。各组均1次/天,连续6周给药。术后每周称体质量,量体长,计算Lee’s指数。实验结束后,检测各组豚鼠血清HDL、LDL、TG、CHO的含量。结果:与模型组相比,鸡血藤提取物可降低去卵巢豚鼠Lee’s指数,从第3周开始明显,其中高剂量效果好。与模型组比较,鸡血藤提取物各组豚鼠血清HDL含量均升高;LDL、TG、CHO含量则均下降,有统计学差异,P<0.05或P<0.01,并呈一定的量效关系,以高剂量效果好。结论:鸡血藤提取物对去卵巢成年动物的脂代谢失衡有调节作用,显示类雌激素样功效,其通过调节血脂水平有预防绝经后妇女因肥胖导致心血管疾病的可能。

Objective： To Explore the effects of the extract of suberect spatholobus stem on lipid metabolism of ovariectomized guinea pig by established model with lipid metabolism disorder of ovariectomized guinea pig. Methods： To establish model with lipid metabolism disorder of ovariectomized guinea pig, thirty female guinea pig were randomly divided into 5 groups （n = 6） ： ①control group;②ovariectomized model group; ③positive drug group （estradiol valerate, 80 mg/kg · d） ; ④low dose of group （ the extract of suberect spatholobus stem, 40 mg/kg） ; ⑤high dose of group （the extract of suberect spatholobus stem, 80 mg/kg） . All ovariectomized guinea pigs must be done continuously checked by vaginal smears method after operation within 1 weeks. After one week following ovariectomized model successfully, the guinea pigs were administrated saline, estradiol valerate or the extract of suberect spatholobus stem by ig once a day for 6 weeks, and measured the body weight and length, then calculated the Lee＇ s index once a week. After the experiment, all guinea pigs serum were took and detected the content of HDL, LDL, TG, CHO. Results： Compared with the model group, Lee＇ s index is decreased in the extract of suberect spatholobus stem groups, high dose of group work better. The content of HDL is raised, and The content of LDL, TG, CHO are decreased in serum of the extract of suberect spatholobus stem groups, high dose of group work better, compared with the model group, there is significantly difference, P 〈0. 05 or P 〈0. 01, and there are positively related to dose and effect. Conclusions： The extract of suberect spatholobus stem can improve adult ani- mals＇ s lipid metabolism disorder caused by ovariectomy, regulate and it can produce the effect with estrogen - like, it can lipid levels, and maybe prevent obese women cardiovascular disease after menopause.
