摘要
为寻找日本血吸虫新基因并进行血吸虫基因功能研究 ,随机挑选日本血吸虫 (中国大陆株 )成虫 c DNA文库克隆 ,培养后提取噬菌体 DNA作模板 ,进行 PCR反应扩增 ,将扩增产物部分测序产生表达序列标签 (EST) ,并将 EST输入Gen Bank和 EMBL,运用分子生物学软件比较其同源性 ,推导其蛋白序列并进行分析 ,结果得到 75个未曾登录的新基因 ,并对其中一些基因进行功能推测研究 ,认为表达序列标签是快速有效寻找基因的方法 。
In order to look for new gene of Schistosoma japonicum, and study function of genes, automated partial DNA sequencing was conducted on randomly selected Schistosoma japonicum (China mainland strain) complementary DNA (cDNA) clones to generate expressed sequence tags (ESTs). Input them to GenBank and EMBL databases to conduct homology analysis and predict probable amino acid sequences and their function. Of the sequences generated, 75 sequences have not been previously described in Schistosoma japonicum, and there are some possible significant biological interest clones. EST is a quick and efficient approach to find new gene. The use of bioinformatics software is promising.
