摘要
目的探讨卵母细胞体外成熟培养液的改良及研究。方法选取昆明小鼠50只作为研究对象,根据培养液的不同将50只小鼠分为A组和B组,每组各25只。A组采用低糖细胞培养基(DMEM)培养,B组采用含15%~20%胎牛血清的人类输卵管液培养,两组在相同条件下进行培养,比较两组的体外受精率、正常受精率、卵裂率、优质胚胎率、可移植胚胎率、妊娠率及活产率。结果A组小鼠的体外受精率(71.4%)、正常受精率(65.7%)及卵裂率(89.1%)与B组(67.6%、68.1%、85.1%)比较,差异无统计学意义(P〉0.05);A组小鼠的优质胚胎率(31.7%)和可移植胚胎率(56.1%)均优于B组(25.0%、42.5%),差异有统计学意义(X2=8.26、10.17,P〈0.05)。A组小鼠的妊娠率(23.3%)低于B组(32.6%),差异有统计学意义(X2=7.58,P〈0.05);但A组小鼠的活产率(17.8%)稍低于B组(18.9%)比较,差异无统计学意义(P〉O.05)。结论低糖DMEM培养液具有较好的应用效果,但仍需进一步的研奔.
Objective To explore the improvement and research of the cultured medium in in--vitro maturation. Meth- ods 50 Kunming mice were selected as the research objects, they were divided into group A and group B according to the different cultured medium, each group of 25 mice. Group A was given low sugar dulbecco's modified eagle medium (DMEM) culture, group B was given human tube fluid containing 15%-20% fetal bovine serum, two groups under the same conditions, the in-vitro fertilization rate, normal fertilization rate, cleavage rate, high quality embryo rate, portable embryo rate and pregnancy rate and live birth rates in two groups were compared. Results The in-vitro fertilization rate (71.4%), normal fertilization rate (65.7%) and cleavage rate (89.1%) in group A of mice compared with group B (67.6%, 68.1%, 85.1%), there were no statistically significant differences (P 〉 0.05), the high quality of mice rates of embryo (31.7%) and portable embryos (56.1%) in group A were better than those of group B, the differences were statistically significant (X2=8.26, 10.17, P 〈 0.05). The pregnancy rate in group A (23.3%) was less than that of group B (32.6%), the difference was statistically significant (X2=7.58, P 〈 0.05), but the live birth rate of mice in group A (17.8%) was little lower than that of group B (18.9%), there was no statistically significant difference (P 〉 0.05). ConcLusion The low sugar DMEM culture has good using effect, but still needs further research.
