摘要
为探讨辐射处理对徐长卿愈伤组织的影响,获得有利变异植株,采用不同剂量60 Co-γ射线处理徐长卿愈伤组织,研究了辐射处理对愈伤组织致死率和分化变异的影响,并对不定芽的分化率和变异率、变异不定芽生根及试管苗移栽和定植进行研究。结果表明:经过剂量为8、12、16和20Gy 4种处理的愈伤组织死亡率随着辐射剂量的提高而增加;经过8、12、16Gy 3种处理剂量处理的愈伤组织能使分化的不定芽发生变异,其变异率随着辐射剂量增加而提高;变异不定芽培养的试管苗生根率、平均生根数、移栽与定植的成活率随着辐射剂量增加而降低。从经8Gy辐射处理的变异试管苗中筛选出1株长势非常旺盛的有利变异试管苗,试管苗的后代生长非常旺盛,有利变异保持不变。
In order to study the effect of radiation treatment on callus of Cynanchum paniculatus and obtain fa- vorable variant plants,using different doses of 60Co-γ rays,the effects of radiation treatment on fatality rate and differentiation variation of callus were analyzed, differentiation rate and mutation rate of adventitious bud, varia- tional adventitious bud rooting, transplanting and engraftment of tube seedlings were studied. The results showed that mortality rate increased with the increasing of radiation dose of 8,12,16,20 Gy on callus ; the 8,12 and 16 Gy three kinds of treatments made differentiation adventitious bud mutation,and the mutation rate im- proved with the increasing of radiation dose. The rooting rate,average root number, survival rate of transplant and engraftment of the variational adventitious bud culture in vitro decreased with the increasing of radiation dose. Screening one strain variational, tube seedlings which was strong and favorable for variation treated with radiation dose 8 Gy. It remained the favorable variation and the offspring tube seedlings grew strong.
