摘要
目的观察β干扰素Toll样受体结构域衔接蛋白(Toll/IL-1 receptor domain containing adaptor inducing IFN-β,TRIF)在Buerger’s病患者腰交感神经节中的表达,探讨TRIF在Buerger’s病发病中的作用机制。方法收集2012年8月至2013年5月我院行腰交感神经节切除术的Buerger’s病患者手术标本30例为Buerger’s病组,同期肝移植供体者腰交感神经节6例为对照组。应用免疫组化、反转录PCR(RT-PCR)技术及蛋白质印迹(Western blot)技术测定2组腰交感神经节中TRIF的表达水平并且比较组间差异。进一步采用激光共聚焦技术,检测TRIF在Buerger’s病患者神经元中的表达特点。结果 Buerger’s病患者腰交感神经节中TRIF的mRNA和蛋白表达水平明显高于对照组(P<0.01)。并且,在Buerger’s病患者腰交感神经节组织中,TRIF主要分布于神经细胞之中。结论 TRIF与Buerger’s病的发生和发展关系密切;Toll样受体信号通路激活可能是Buerger’s病发病的重要因素。
To observe the expression and distribution of Toll/IL-1 receptor domain-containing adaptor inducing IFN-β (TRIF) in the lumbar sympathectomy of Buerger's patients, resections of lumbar sympathetic ganglion from 30 patients with Buerger's disease were analyzed, with 6 cases of donors for hepatic transplantation as control group. The expression of TRIF was detected by immunohistochemistry, reverse transcription quantitative PCR (RT-PCR) and Western blot technology in Buerger's disease group and control group. In Buerger's disease group, the results of RT-PCR and Western blot indicated that the expression of TRIF was much higher than that of in control group (P〈 0.01). Immunohistochemistry and immunofluorescence staining images revealed that TFIF was mainly expressed and distributed in the nerve cells of lumbar sympathetic ganglia tissues. Our results indicated that the expression of TRIF is significant increased in lumbar sympathetic ganglion of 30 patients with Buerger's disease, and the activation of TLRs-TRIF may correlated with the occurrence and development of Buerger's disease.
