体检人群蛋清IgG抗体检测及不耐受组分分析 被引量:5

摘要 目的分析体检人群蛋清IgG抗体水平,探讨蛋清不耐受与年龄、性别的关系,并进行不耐受组分分析。方法碳酸氢钠溶解法制备蛋清提取物,并用粗提蛋白包被微孔板,采用酶联免疫吸附法检测391例健康体检者蛋清IgG水平,分析蛋清不耐受与年龄、性别的关系;电泳分析其蛋白组分,并用酶免疫印迹方法分析提取液中与IgG特异性结合的主要蛋白组分。结果本地区体检人群蛋清不耐受抗体阳性率为58.82%,其中女性(65.50%)显著高于男性(48.54%),差异有统计学意义(P<0.05);在人群中不同年龄组的不耐受率不同,且差异有统计学意义(P<0.05);酶免疫印迹结果显示,蛋清中不耐受组分主要有4种,且个体间所针对的抗原无差异。结论体检人群中普遍存在蛋清IgG抗体,且鸡蛋不耐受可能与年龄和性别相关;不同个体间蛋清主要不耐受组分无差异。 In this paper, we aim to detect and analyze the level of albumen-specific IgG in health examination population, and to investigate the relationship of albumen intolerance with age and gender, and to explore the main intolerance component in albumen. Firstly, the albumen protein was extracted using sodium bicarbonate dissolve method, and the microplates were coated with the crude protein. The albumen-specific IgG were detected with enzyme linked immunosorbent assay (ELISA) in 391 healthy persons and analyzed the relationship of albumen intolerance with age and gender. The component of crude protein was analyzed by electrophoresis and the main intolerance ingredients were identified by Western-blotting. Data showed the positive rate in the cohort was 58.82%, and positive rate in females (65.50%) was higher than those in males (48.54%) (P〈 0.05); the intolerance rate was different in different age groups (P 〈 0.05). Western-blotting all showed there were four main intolerance components in the albumen, and different individuals showed no difference when response to these compnents. The results indicated that the albumen-specific IgG antibodies exist commonly in health examination group; age and gender might be associated with egg intolerance, while there are no difference in the main intolerance components between individuals.
