摘要
目的:通过观察维持性血液透析(Maintainance hemodialysis,MHD)患者体内白细胞介素17(Interleukin17,IL-17)和白细胞介素23(Interleukin23,IL-23)水平、相互之间的相关性,探讨IL-17和IL-23的水平变化及其临床意义。方法:采集10例健康体检者作为正常对照组,20例慢性肾衰竭终末期新进入透析患者的外周血。酶联免疫吸附法(ELISA法)检测所有血清样本的IL-17和IL-23水平,逆转录-聚合酶链反应(RT-PCR)检测外周血淋巴细胞中IL-23mRNA表达水平。结果:(1)MHD患者血清IL-17、IL-23以及外周血淋巴细胞中IL-23mRNA水平明显高于正常对照组(P<0.01),且第16次透析前IL-17、IL-23、IL-23mRNA表达高于第4次透析前(P<0.01);(2)MHD患者血清IL-17表达和IL-23、IL-23mRNA表达呈正相关(r=0.743,P<0.01;r=0.575,P<0.01)。结论:IL-17和IL-23的高表达可能参与了MHD患者的慢性炎症形成过程,两者表达水平有密切相关性。
Objective:To observe the changes of IL-17 and IL-23 in patients with maintainance hemodialysis (MHD) for exploring the relation between them and clinical significance Methods:Ten sera from normal people and twenty sera from patients hemodialyzed for more than one week were sampled.Serum IL-17 and IL-23 were measured in all the sera with sandwich ELISA.Reversing transcription-polymerase chain reaction (RT-PCR) detection of IL-23mRNA expression in peripheral blood lymphocytes.Results:(1)Serum IL-17,IL-23 and IL-23mRNA in patients were much higher than those in normal group(P < 0.01),and serum IL-17,IL-23 and IL-23mRNA before 16th hemodialysis were higher than those before 4th hemodialysis (P <0.01).(2)IL-17 was positively correlated to IL-23 (r =0.743,P < 0.01) and IL-23mRNA (r =0.575,P < 0.01).Conclusion:IL-17 and IL-23 in patients is much higher than that in normal people,IL-17 are positively correlated to IL-23,IL-23mRNA.Suggesting that IL-17 and IL-23 play an important role in pathogenesis of immune anomaly and micro-inflammation in patients with MHD.
