摘要
目的 探讨 p16基因对人膀胱癌细胞周期的影响 .方法 通过脂质体介导的基因转移技术将野生型 p16基因导入人膀胱癌细胞系 T2 4中 ,绘制细胞生长曲线 ,用流式细胞仪检测转染前后细胞周期的变化 .结果 与对照组相比 ,转染p16基因的膀胱癌细胞生长速率降低 ,流式细胞仪检测显示
AIM To elucidate the inhibitory effects of p16 on human bladder carcinoma cell cycle. METHODS Wild type p16 was transfected into human bladder carcinoma cell line T24 by using Lipofectin mediating genetransfer technique. The cell growth curve was drawn. Cell cycle was detected by flow cytometry (FCM) before and after p16 transfection respectively and the change was compared. RESULTS Compared with the controls, the growth rates of p16 transfected cells were greatly decreased. FCM analysis showed that S phase fraction and proliferation index were reduced obviously. CONCLUSION P16 could inhibit DNA synthesis of tumor cell by preventing the transition from G to S, which exerts obvious inhibitory effects on bladder carcinoma cell growth.
