乳铁蛋白对大鼠成骨细胞增殖及IGF-1/IGF-1R mRNA表达的影响 被引量:8

摘要 目的探讨乳铁蛋白对大鼠成骨细胞增殖及胰岛素样生长因子1(IGF-1)/和胰岛素样生长因子1受体(IGF-1R)表达的影响。方法取出生24 h内SD大鼠6只,无菌条件下取出颅骨,用混合酶消化法进行成骨细胞培养,用不同浓度乳铁蛋白(0、0.1、1、10、100、1 000μg/mL)干预成骨细胞1、3、5、7 d后,用噻唑蓝比色法(MTT)测定成骨细胞增殖,用实时荧光定量聚合酶链反应(real time-PCR,RT-PCR)检测乳铁蛋白对IGF-1及IGF-1R mRNA表达的影响。结果 MTT结果显示与对照组相比,浓度为1、10、100、1 000μg/mL的乳铁蛋白在不同时间均能促进成骨细胞增殖,差异有统计学意义(均P<0.05)。随时间推移,不同浓度乳铁蛋白均可使成骨细胞数量增加,其中100μg/mL组第7天细胞增殖数达到最大。RT-PCR结果显示,与对照组相比,浓度为10、100、1 000μg/mL的乳铁蛋白在各时间段均能增加成骨细胞IGF-1/IGF-1R mRNA的表达量,差异有统计学意义(均P<0.05),且随时间推移呈增加趋势,上述浓度乳铁蛋白对成骨细胞IGF-1/IGF-1R mRNA的表达量在第5、7天差异有统计学意义(均P<0.01)。结论乳铁蛋白能促进大鼠成骨细胞增殖,促进IGF-1/IGF-1R mRNA表达,可能是乳铁蛋白防治骨质疏松的机制之一。 goes on , the number of osteoblasts were increased exposed to different concentrations of lactoferrin , the number of osteo-blasts got the peak at the 100μg/mL in the 7th days.The IGF-1/IGF-1R mRNA expression of the osteoblasts exposed to 10, 100, 1 000 μg/mL lactoferrin were higher compared with the control group in each time (P&lt;0.05), The IGF-1/IGF-1R mRNA expression of the osteoblasts exposed to various concentration of lactoferrin were significantly higher com -pared with the blank control group in 5, 7day (P&lt;0.01).Conclusion Lactoferrin can increase the proliferation and the IGF-1/IGF-1R mRNA expression in rat primary osteoblasts .It may be one of the mechanism of lactoferrin prevention and cure of osteoporosis .
