脑缺血再灌流后脑组织一氧化氮水平变化及其与细胞凋亡的关系 被引量:23

摘要 目的 探讨一氧化氮 (NO)在局灶性脑缺血再灌流损伤中的作用。方法 采用大鼠线栓法大脑中动脉闭塞 (MCAO)模型 ,观察脑缺血 2h再灌流后 2、 6、 18、 2 4及 48hNO含量动态变化 ,同时应用流式细胞仪检测DNA断裂百分率 ,并分析二者之间的关系。结果 脑缺血 2h再灌流后 6hNO水平开始升高 ,随再灌流时间的延长 ,其升高更明显 ;再灌流 18h后DNA断裂百分率明显升高 ,其变化趋势与NO相似。结论 上述结果提示 ,脑缺血再灌流后过量产生的NO与再灌流后神经细胞凋亡的发生有关。 Objective To clarify the mechanisms of neurotoxic effect of NO following experimental focal cerebral ischemic reperfusion.Methods NO contents and DNA fragmentation were assessed in rat ischemic brain area following 2 hour MCAo.Results The contents of NO and DNA fragmentation increased proportionally within 48h reperfusion.Conclusions Our results suggest that excessive formation of NO plays an important role in neuronal apoptosis.
