人脑胶质瘤免疫抑制因子TGFβ_2及TGFβ_1的基因表达 被引量:2

摘要 目的 研究人脑胶质瘤主要免疫抑制因子转化生长因子TGFβ2 及TGFβ1的基因表达与其胶质瘤恶性程度的关系。方法 采用Northern杂交、免疫组化和Western杂交法检测了 5 0例胶质瘤、3例恶性胶质瘤体外细胞系和 8例正常脑组织TGFβ2 的表达水平。同时检测其同型异构体TGFβ1的表达水平。结果 正常脑组织几无TGFβ2 和TGFβ1表达 ,而胶质瘤均表达 2 .8和 /或 5 .1kb的TGFβ2 mRNA片段和约 2 5 0 0 0u及 30 0 0 0u的蛋白 ;TGFβ2 和TGFβ1表达水平随肿瘤恶性程度增高而增高。结论 TGFβ2 和TGFβ1表达水平与肿瘤恶性程度呈正相关 ,TGFβ2 和TGFβ1可作为恶性胶质瘤免疫基因治疗的候选基因。 Objective To Study the gene expression of Transforming growth factorβ 2(TGFβ 2) and TGFβ 1 in gliomas. Methods TGFβ 2mRNA and protein level as well as TGFβ 1 protein level in 50 specimens of gliomas with different grade of malignancy,3 malignant human glioma cell lines and 8 normal brain tissues were examined by Northern blot, immunohistochemistry and Western blot analysis. Results Expression of 2.8 kb and/or 5.1 kb mRNA was found in all tumors with different grade of malignancy, and expression of 25 000 u and 30 000 u protein was also found, but there was no expression of TGFβ 2 and TGFβ 1 in normal brain tissues.On the mRNA and protein level, there was significant difference among Ⅲ,Ⅳ grade, cell lines and low grade gliomas. Conclusions The expression of TGFβ 2 and TGFβ 1 was positively correlated with the tumor grade, so they could be a good target for immuno gene therapy of malignant gliomas.
