摘要
[目的]确定FMDV抗原的最佳纯化方法及纯化条件,为其在我国动物用疫苗行业中的大规模应用和推广奠定基础。[方法]选用大孔树脂和离子交换填料浓缩纯化FMDV抗原,通过对样品及缓冲液的pH、流速、离子强度、上样量等方面探讨各填料的最佳工作参数,并比较2种方法浓缩纯化同种FMDV抗原的效果。[结果]大孔树脂处理样品的抗原回收率为21%,离子交换填料的抗原回收率为11.7%,均不理想。大孔树脂的杂蛋白去除率为90%,离子交换填料的杂蛋白去除率仅为52%,说明大孔树脂在去除抗原中杂蛋白的效果方面可达到所需的要求。[结论]这2种方法浓缩纯化FMDV抗原的效果均不理想,但大孔树脂效果稍好。
[ Objective] The research aimed to discuss the best purificati0n methods and purification conditions of FMDV antigen and lay the foundation for its large-scale application and popularization in the animal bacterin industry in China. [ Method] The macroporons resin and ion exchange paddings were selected to concentrate and purify FMDV antigen. The optimum working parameters of different paddings were dis- cussed from pH,flow rate, ionic strengthen,sample volume and other aspects of samples and buffer solution. The effects of concentrating and purifying FMDV antigen by these two methods were compared. [ Result] The antigen recovery rate of samples treated with macroporous resin was 21% and that of ion exchange paddings was 11.7% , which were both not ideal. The removal rate of impurity protein with maeroporous res- in was 90% and that with ion exchange paddings was only 52% , which indicated that macroporous resin could meet the demands for removing impurity protein from antigens. [ Conclusion] The effects of concentrating and purifying FMDV antigen by these two methods were not ideal, and the effect of macroporous resin was better.
