2型糖尿病大鼠骨髓干细胞条件培养基促进糖尿病大鼠脑梗死后神经功能恢复 被引量:1

摘要 目的 探讨2型糖尿病(T2DM)大鼠骨髓干细胞条件培养基(BMSC-CM)对T2DM大鼠脑缺血/再灌注损伤后神经功能恢复的影响.方法 将成年雄性SD大鼠用烟酰胺、链脲佐菌素(STZ)诱导制作T2DM模型,制备T2DM大鼠BMSC-CM,线栓法制作T2DM大鼠大脑中动脉栓塞(MCAO)脑缺血/再灌注模型.实验动物分为空白对照组、单纯培养基组和条件培养基治疗组(每组n=6).治疗组分别于术后24、48 h经尾静脉注射T2DM大鼠BMSC-CM(10 ml/kg),单纯培养基组经尾静脉注射培养基DMEM,空白对照组未给予任何干预措施.术后1、7、14天分别进行神经功能缺损评分(mNSS)和足错步试验.结果 术后第1天,各组mNSS、足错步次数比较差异无统计学意义(P>0.05).术后第7、14天,条件培养基治疗组mNSS、足错步次数明显低于空白对照组和单纯培养基组(P<0.05),空白对照组与单纯培养基组mNSS、足错步次数差异无统计学意义(P>0.05).结论 T2DM大鼠BMSC-CM可明显促进T2DM大鼠脑梗死后的神经功能恢复. Objective To investigate the protective effects of type 2 diabetes mellitus (T2DM) rat - derived bone marrow stem cells - conditioned medium ( BMSC - CM) on neurological function of T2DM rats with cerebral ischemia/ reperfusion injury. Methods T2DM was induced in adult male SD rats by injecting nicotinamide and streptozotocin (STZ). T2DM rat -derived BMSC -CM was prepared r these T2DM rats. T2DM rats were subjected to middle cere bral artery occlusion (MCAO) with a nylon monofilament suture and to establish cerebral ischemia/reperfusion model. The rats were assigned to three groups (n = 6 each). The treatment group was treated with T2DM rat - derived BMSC - CM at 24 and 48 h after MCAO surgery. The culture medium group was treated with DMEM at 24 and 48 h after MCAO surgery. The blank control group did not receive any intervention. Behavioral test (Foot Fault test) and modified neuro- logical severity score (mNSS) were performed before and at 1,7, 14 days following MCAO surgery. Results There was no significant difference in the mNSS and the number of foot fault among the three groups at 1 day following MCAO sur- gery (P 〉 0.05 ). There was also no significant difference in mNSS and the number of foot fault between the blank control group and culture medium group at 7 and 14 days following MCAO surgery ( P 〉 0.05 ). Compared with the blank control and culture medium group, the mNSS and number of foot fault improved significantly at 7 and 14 days following MCAO surgery in the treatment group ( P 〈 O. 05 ). Conclusion T2DM rat - derived BMSC - CM can significantly improve neu- rological function in T2DM rats after stroke.
