金叶马褂木叶片特性及其RAPD分子鉴定

Leaves characteristics and RAPD molecular identification of tulip tree with golden leaves

金叶马褂木是从当地金边马褂木中发现的春季至初夏间全叶呈金黄色至淡黄色的芽变材料。对该芽变种质的叶片特性进行了连续8年的观察与调查,同时结合RAPD技术进行了初步的分子鉴定。叶片特性观察与调查结果表明,金叶马褂木叶片大小与金边马褂木、北美马褂木差异不大,但叶片叶绿素含量显著少于金边马褂木和北美马褂木,叶片色泽也与金边马褂木有显著性差异。结合RAPD分子鉴定结果,认为金叶马褂木是一个发生变异的新种质。

Tulip tree with golden leaves is bud mutation material, its leaves color changes from golden yellow in the spring to pale yellow in early summer. In this paper, main leaves characteristics of the bud variant in eight successive years were observed and inves- tigated. Meanwhile, RAPD technique was used to perform the preliminary molecular identification. Leaves observation results showed that the leaves size of tulip tree with golden leaves was not different from those of tulip tree with golden leaves ledge and North American tulip tree, while chlorophyll content of the former tulip tree was significantly less than that of others. Meanwhile, it was also found that leaves color changes of tulip tree with golden leaves were significant different with that of golden leaves ledge. Combining with RAPD molecular identification results, it is judged that tulip tree with golden leaves is new germplasm with variation.
