摘要
目的检测新的长链非编码RNA(LncRNA)T-ALL-R-LncR1在胚胎型横纹肌肉瘤(ERMS)中的表达。方法对BALB/c裸鼠皮下注射RD细胞悬液建立ERMS动物模型;成瘤后肿瘤组织行HE染色和免疫组织化学检测,镜下观察结果。RT-PCR法检测T-ALL-R-LncR1在RD细胞和ERMS动物模型肿瘤组织中的表达。结果接种RD细胞的裸鼠全部出现移植瘤,成瘤率100%。镜下观察HE染色及免疫组织化学切片后确定肿瘤组织病理分型为ERMS。通过RT-PCR检测及测序在RD细胞和ERMS动物模型肿瘤组织中发现T-ALL-R-LncR1。结论成功建立ERMS动物模型。新的LncRNA T-ALL-R-LncR1可能为今后研究ERMS的诊断及治疗提供了新的靶标。
Objective To investigate the expression of long non-coding RNA (LncRNA) T-ALL-R-LncR1 in RD cells and tumor tissues of embryonal rhabdomyosarcoma (ERMS). Methods The suspension of RD cells was injected into BLAB/c nude mice to establish ERMS model. And then the tumor tissues were observed under microscope by HE staining and immunohistochemistry. The expressions of T-ALL-R-LncR1 in RE) cells and tumor tissues of ERMS mouse model were detected by RT-PCIL Results Solid xenografts were obviously seen in all nude mice after implantation of RD cells. HE staining and immunohistochemistry confirmed that the pathology classification of the tumor tissues was ERMS. T-ALL-R-LncR1 was found in RD cells and tumor tissuses of ERMS mouse model by RT-PCR and sequencing. Conclusion ERMS mouse model has been successfully established. LncRNA T-ALL-R-LncR1 may be a new target for the diagnosis and treatment of ERMS.
