摘要
以紫花亚菊(Ajania purpurea)组培苗茎段、叶片为外植体,在附加不同浓度植物生长调节剂6-BA和NAA的MS培养基上诱导培养,获得再生植株。对试验结果进行观察分析筛选出合适的配方:愈伤组织诱导培养基为MS+1.0 mg·L-16-BA+0.1 mg·L-1NAA,诱导率达95.6%;芽诱导培养基为:MS+1.0 mg·L-16-BA+0.2 mg·L-1NAA,诱导率达82%;不定根最适诱导培养基为:1/2 MS+0.15 mg·L-1IBA,生根率达90%以上,再生植株移栽成活率达90%。
A plant regeneration system from leaves and stems of tissue-cultured Ajania purpurea was developed in this study.The results showed that the callus was induced with a rate of 95.6%on an optimal medium MS+1.0 mg·L-1 6-BA+0.1 mg·L-1 NAA.The highest induction rate of the adventitious shoots from the calli on the medium of MS+1.0 mg·L-1 6-BA+0.2 mg·L-1 NAA was 82%.The optimal medium for adventitious root inducing was 1/2MS+0.15 mg·L-1 IBA, with a rooting rate of 90%.The plantlets were transplanted to pots with sands , vermiculite and peat to acclimate for 2 -3 weeks.Then the survival plantlets were transferred outdoors with 90% transplanting survival rate.
