摘要
何首乌用70%甲醇超声提取浓缩后,用氯仿萃取分离出游离和结合型蒽醌,得到定量用试液。在439nm检测波长下,以甲醇:异丙醇:水=80:10:10(磷酸调pH值为3.0)为流动相,C18填料为固定相,采用HPLC法测定何首乌中蒽醌类成分含量。平均加样回收率大黄素为100.7%,RSD=1.04.%大黄素甲醚为98.7%,RSD=1.77%% 重复性实验的RSD分别为:大黄酸1.18%,大黄素0.79%,大黄素甲醚0.75%(n=10)。
Using 70% methanol, the Polygonum multiforum extract was concentrated by ultrasonic method .Then concentrated extract was leached with chloroform and separated into free and bound anthraquinones.The mobile phase was V(CH3OH):V(C3H7OH):V(H2O)=80:10:10 (control pH to 3.0 with phosphorus acid ).The HPLC system was consisted of Hypersil BDS C 18(5μm,250nm×46nm) and UV detector set at 439nm.The experiment results show that the average recovery of Emodin was 100.7% (RSD=1.77),and that of Emodin methylether was 98.7% (RSD=1.77%) .The relative standard deviation for Emodin acid ,Emodin and Emodin methylether were 1.18%,0.79% and 0.75% respectively.
