大球盖菇原生质体再生及单核化特性的研究 被引量:20

摘要 大球盖菇原生质体再生条件及单核化特性结果。原生质体再生速度极快,涂布平板3d后肉眼即可见明显的再生菌落形成,在PGPM再生培养基上再生率为0.97~2.0%,渗稳剂种类对再生率无明显影响,但可影响再生菌落形态,液体预培养1~2d,再生率明显下降;大球盖菇原生质体单核化率高达77.6%,且再生双核体和再生单核体在形成再生菌落时无时间差,其生长速度亦无快慢之分,液体预培养可显著减少单核化率,再生单核体中存在亲本两种交配型,但二者的比率不为1。 Regeneration and monokaryotation of Stropharia rugoso-annulata protoplasts were investigated. The results showed that the protoplast regeneration was very fast, and the regenerated colonies could be seen by naked eyes after plating 3d. The regeneration rate was 0.97~2.00% on PGPM. The osmotic stabilizers of mannitol, sucrose and magnesium sulphate had no significant effects on regeneration rate, but could affect the morphology of regenerated colonies. Pre-culture of protoplasts in liquid regeneration media for 1 --2d at 25 C could decrease the regeneration and monokaryotation rate. The protoplast monokaryotation rate was 77.7%. The regenerated dikaryons and monokaryons could be formed on the plates simultaneously, and had no difference in mycelium growth rate and colony-forming time. Two kinds of parent mating types could be found among protoplasted monokaryons, but the ratio was not 1: 1. It was the first report about the regeneration and monokaryotation characters of Strapharia of Strapharia rugoso-annulata.
