细胞与病毒同时接种于 33℃培养 5天 ,其病毒滴度平均为 6 2 5LogCCID50 /ml,但细胞成片后再感染 ,其病毒滴度平均为 4 85LogCCID50 /ml。在鸡胚细胞感染 2 .3× 10 -3 病毒剂量时 ,滴度可达≥ 6 5LogCCID50 /ml,病毒高峰期于 10 0小时和 12 0小时分二次收获 ,其滴度分别达到 6 0和≥ 6 5LogCCID50 /ml。每瓶产疫苗量 45 0 0ml和90 0 0ml,病毒滴度没有影响。液体疫苗于 2~ 8℃保存 14天 ,滴度由 5 75降至 4 75LogCCID50 /ml,而将疫苗保存在 - 18℃至 18周 ,其滴度由 6 38缓慢下降至 5 0LogCCID50
The optimal condition of preparation of mumps vaccine with 15 L rolling bottle is reported.The mean titers were 6.25 LogCCID 50 /ml when the cells and virus were simutaneously inoculated and incubated at 33℃ for five days.After the cell monolayer were formed,which were infected by the virus,the mean titer were 4.85 LogCCID 50 /ml.The chick embryo cells were infected with the virus at multiplicity of infection(MOI)of 2.3×10 -4 the virus titer may reach to 6.5 LogCCID 50 /ml.The virus suspension were harvested two times at 100 h and 120 h at the peak of virus titers,the titer of them were 6.00 and >6.50 LogCCID 50 /ml respectively.The yield of the vaccine were 4500 ml and 9000 ml per bottle without significant changes in their titers.The vaccine in liquid state were stored at 2 10℃ for 14 days and the titer had dropped from 5.75 to 4.75 LogCCID 50 /ml.While the vaccine was stored at -18℃ and the titer had slowly declined from 6.38 to 5.00 LogCCID 50 /ml.
