人喉上皮细胞癌细胞系Hep-2高分子DNA的提取及检测

Extraction of the High Molecular Weight DNA of An Epidermoid Carcinoma of Human Larynx Line Hep-2

本实验分別提取了三批Hep-2细胞高分子DNA,并用紫外分光光度计测定其纯度和含量,用琼脂糖凝胶电泳检测.在提取过程中,用化学方法代替RNA酶水解RNA,结果在DNA琼脂糖凝胶电泳图上未见RNA区带.

Three groups of the high molecular weight DNA of the epidermoid caroinoma of human larynx line Hep-2 were extracted. The purity and content of the extracted DNA were assayed by the ultraviolet spect rophotometer and egarose gell electrophoresis. In the course of extraction the RNA of cell could be hydrolyzed by using a chemical way instead of RNase, and the result showed that the band of RNA was not shown on the elecrtrophoresis photograph.
