摘要
目的 探索PGE2 抑制瘢痕成纤维细胞胶原合成的机理。方法 从新鲜的增生期瘢痕组织中培养成纤维细胞 ,采用Dotblot杂交法观测PGE2 对瘢痕成纤维细胞Ⅰ、Ⅲ型前胶原基因表达的影响。结果 PGE2 显著降低瘢痕成纤维细胞内Ⅰ、Ⅲ前胶原mRNA的含量。结论 PGE2 在胶原转录水平抑制瘢痕成纤维细胞的胶原合成。
Objective The mechanisms of inhibitory effects of PGE 2 on collagen synthesis in scar derived fibroblasts was investigated.Methods Fibroblasts was isolated from fresh hypertrophic tissue and cultured, exposed to PGE 2. The concentration of type Ⅰ and Ⅲ procollagen mRNA expression in scar-fibroblasts were assayed by Dot blot hybridization. Results PGE 2 significantly reduced type Ⅰ and Ⅲ procollagen mRNA expression in scar-fibroblasts. Conclusion PGE 2 inhibits collagen synthesis of scar-fibroblasts at transcriptional level.
