摘要
目的:探讨阿魏酸对颅脑损伤的小鼠脑组织的保护作用。方法:50只小鼠随机分成正常组、模型组、阿魏酸低剂量组(40 mg·kg-1·d-1)、阿魏酸中剂量组(80 mg·kg-1·d-1)、阿魏酸高剂量组(160 mg·kg-1·d-1),每组10只小鼠。采用自由落体法建立小鼠颅脑损伤模型,伤后3 d给药组小鼠灌胃阿魏酸溶液,正常组和模型组灌胃等体积的生理盐水。连续用药7 d后处死小鼠,采用HE染色法及尼氏体染色法观察小鼠颅脑损伤神经细胞形态改变,利用干湿重法测定脑组织含水量,及检测各组颅脑损伤小鼠脑组织NO含量和线粒体中三磷酸腺苷(ATP)酶活性。结果:颅脑损伤后,阿魏酸组小鼠脑组织病理改变减轻,NO含量及脑组织含水量较模型组显著降低,ATP酶活性较模型组明显增加。结论:阿魏酸能够有效减轻颅脑损伤小鼠的病理改变,提高损伤后脑组织中ATP酶活性,减少NO产生,改善脑水肿,保护颅脑损伤的小鼠。
OBJECTIVE To investigate the protective effect of ferulic acid (FA) on the mice with craniocerebral injury. METHODS Fifty mice were randomly divided into 5 groups as following: normal group, model group, low dose FA treated group(40 mg·kg^-1·d^-1 ) ,medium dose FA treated group(80 mg·kg^-1·d^-1 ) ,high dose FA treated group(160 mg·kg^-1·d^-1 ), 10 mice in each goup resecptively. The mice of craniocerebral injury model were established with free failing method. The trea- ted group was administrated with oral FA for 7 days after three days of model reproduction, while the normal group and model group treated with normal saline. The mice were sacrificed after 7 days treatment. The changes of neuronal cells were detected by hemotoxylin and eosin staining, and Nissle staining, and the contents of water in brain tissue were measured by wet-dry weight. The nitric oxide(NO)contents and ATP activity in brain tissues were determined by commercial kit. RESULTS After the treatment, compared with the model group, the pathological changes of brain tissues were significantly alleviated in treated group, and the ATP activity was increased, and the level of NO and water content were reduced obviously. CONCLUSION Ferulic acid can effectively reduce the pathological changes in mice with craniocerebral injury, enhance ATPase activity in the cerebral tissue damage, reduce the concentration of NO, decrease the cerebral edema, and finally protect the brain against craniocerebral injury.
