摘要
采用菌落直径离体测定法测定盐酸小檗碱对19种植物病原真菌的菌丝生长抑制作用并探讨其抑菌机理.以小麦赤霉菌为例,通过显微镜观测盐酸小檗碱对病原真菌菌丝形态结构的影响;利用电导率法测定盐酸小檗碱对细胞膜通透性的影响及对病原真菌的物质吸收、代谢过程的影响.结果表明,不同质量浓度的盐酸小檗碱均具有广谱抑菌活性.在药物胁迫下,小麦赤霉菌菌丝生长受到了明显抑制,电导率增加,细胞膜通透性降低,细胞膜结构、功能的稳定性遭到破坏.同时,盐酸小檗碱还影响小麦赤霉菌菌丝的还原糖吸收利用,但对琥珀酸脱氢酶的比活力影响不大.
The anti-fungus spectrum of berberine hydrochloride was studied by measuring the fungus colony diameter growth. Fusarium graminearum was selected for the studies of anti-fungus physiologi- cal mechanism of berberine hydrochloride. The cell membrane conductivity and microscopic structure were studied to test the effect of berberine hydrochloride on pathogenic fungi material absorption and metabolism process. The results showed that berberine hydrochloride had a broad spectrum of anti-fun- gi activity. Under the stress of drug, fungus mycelium growth was obviously inhibited, cell conductivity increased and cell membrane permeability was reduced, which indicates the cell membrane structure and function stability of fungus were destructed. In addition, berberine hydrochloride can affect the sugar absorption of the fungus cell and decrease the activity of succinate dehydrogenase slightly.
