摘要
目的:观察运动对糖尿病大鼠心肌肌浆网Ca2+调控蛋白基因表达的影响,探讨运动对心脏的保护作用的分子机制。方法:4月龄雄性SD大鼠,分为安静对照组、安静糖尿病和运动糖尿病组。运动组进行8周耐力训练(20 m/min,60 min,坡度0,每周5次)。检测心肌肌浆网钙调控蛋白Ca2+ATP酶(SERCA2)、兰尼碱受体II型(RyR2)及受磷蛋白(PLB)mRNA表达水平。结果:安静糖尿病组SERCA2和PLB的mRNA表达上调,RyR2 mRNA表达下调,运动使糖尿病组SERCA2和PLB的mRNA表达下调,RyR2 mRNA表达上调。结论:8周耐力运动逆转了糖尿病大鼠心肌肌浆网钙调控蛋白基因表达的改变,提示耐力运动具有预防或逆转糖尿病型心肌病变的作用。
The purpose of this study was to explore the influence of exercise on gene expression of cardiac calcium handling protein in diabetic rats, and to detect the molecular mechanism of the protective effect of exercise on heart. Methods: Four-month old Sprague-Dawley rats were divided into three group randomly: sedentary control group (SC), sedentary diabetic group (SD) and exercise diabetic group (ED). The rats in ED group participated in 8 weeks training(20 rn/min, 60min/day, grade 0°, 5 times/week). The mRNA expressions of Ca2+ -ATPase (SER- CA2), type 2 ryanodine receptors (RyR2), and phospholamban (PLB)were measured. Results: The mRNA expressions of SERCA2 and PLB in SD group increased while that of RyR2 decreased. After exercise, the mRNA expressions of SERCA2 and PLB in ED group decreased while that of RyR2 increased. Conclusion : Eight weeks endurance exercise reversed the gene expressions of calcium handling proteins of myocardial sarcoplastic reticulum in diabetic rats, which suggested that endurance training could prevent or therapy the diabetic cardimyopathy.
