内耳免疫反应中白细胞上α4-整合素和L-选择素的表达

Expression of α4 integrin and L selectin on the surface of leukocytes during inner ear immune response

目的 探讨α4 整合素 (α4 integrin)和L 选择素 (L selectin)在内耳免疫反应中的作用。方法 采用钥孔虹戚血蓝蛋白激发已 2次全身致敏的大鼠内耳 ,诱发其内耳免疫反应 ,制造迷路炎模型 ,然后通过免疫组织化学技术 ,检测内耳L selectin在内耳中的表达。同时用流式细胞仪检测L selectin和α4 integrin在外周血白细胞上的表达。用ELISA法检测外周血白细胞介素 1(IL 1)浓度。结果 内耳炎症期间L selectin表达与炎症程度成反比 ,α4 integrin的表达上调。迷路炎期间 ,IL 1浓度明显升高。结论 内耳免疫反应时 ,α4 integrin在炎性细胞从循环系统进入内耳的过程中发挥着重要作用。α4 integrin升高与外周血IL

Objective To investigate the role played by α4 integrin and L selectin in the immune response during inner ear inflammation. Methods Labyrinthitis was induced in rats by inoculation of keyhole limpet hemocyanin(KLH) into the scalae tympani of the animals that had been systemically sensitized. Expression of L selectin was examined with monoclonal antibodies to L selectin with immunohistochemistry. At the same time, ELISA was used to monitor the interleukin (IL) 1 in the peripheral blood of these animals. In addition, the expression of α 4 integrin and L selectin on the surface of leukocytes in peripheral blood were examined with flow cytometry. Results Expression of L selectin was not found on the surface of leukocytes during the inner ear inflammation. By flow cytometry, the expression of L selectin on the surface of leukocytes in peripheral blood showed down regulated during inflammation. In contrast, the expression of α4 integrin on the surface of leukocytes in peripheral blood showed up regulated during inflammation. The concentration of IL 1 in peripheral blood increased during inner ear inflammation. Conclusions The study further elucidated that the adhesion molecules played on important role on extravasation of inflammation cells from blood vessels during inner ear immune response. And, the increased expression of α4 integrin had related closely to the concentration of IL 1 in peripheral blood during inner ear inflammation.
