摘要
建立了液相色谱-串联质谱法(LC-MS/MS)快速测定烟草中18种游离氨基酸的分析方法。结果表明,18种氨基酸的检出限(S/N=3)为0.5×10-4~0.1mol/L,线性相关系数均大于0.9990,峰面积测定的相对标准偏差(RSD)为0.42%~8.09%,低、中、高3个添加水平的平均回收率为90.0%~106.0%。该方法准确、灵敏,可用于烟草样品中氨基酸的分析检测。
A method for fast analysis of 18 amino acids in tobacco was developed using liquid chromatography- tandem mass spectrometry(LC-MS/MS). The results showed that the limits of detection(LODs) of 18 amino acids were in the range of 0.5 × 10 -4-0.1 mol/L(S/N= 3), the linear correlation coefficients were above 0. 9990 in all cases, and the relative standard deviations(RSDs) of peak areas of the extracted ion chromatograms were in the range of 0.42 %-8.09 %. The average recoveries of the target compounds spiked in the sample at three concentration levels were in the range of 90.0%-106.0%. The developed method is of good efficiency, sensitivity and selectivity, and can be successfully used to determine amino acids in various tobacco samples.
