原发性胆汁性肝硬化患者外周血CD4^+CD25^+Foxp3^+调节性T细胞检测及其临床意义 被引量:7

摘要 目的探讨检测原发性胆汁性肝硬化(PBC)患者外周血CD4+CD25+Foxp3+调节性T淋巴细胞的水平与意义。方法采用流式细胞术检测32例PBC患者和23例健康体检者外周血CD4+CD25+Foxp3+调节性T淋巴细胞亚群比例;采用实时荧光定量聚酶链反应(PCR)技术检测人叉头型基因p3(Foxp3)mRNA表达的水平。结果PBC患者外周血CD4+CD25+Foxp3+调节性T淋巴细胞明显低于对照组(P<0.05);Foxp3的mRNA水平差异无统计学意义(P>0.05)。结论 PBC患者外周血CD4+CD25+Foxp3+调节性T淋巴细胞减少,免疫抑制功能减弱,提示CD4+CD25+Foxp3+调节性T淋巴细胞在PBC的免疫发病机制中可能起重要作用,对评估PBC患者细胞免疫状态和预后有重要价值。 Objective To investigate the roles of CD4+ CD25 + Foxp3 + regulatory T cells (Treg)in the primary bili-ary cirrhosis patients.Methods The flow cytometry was used to evaluated the population of CD4+ CD25 + T cells in the peripheral blood of the PBS patients.Forkhead/winged helix transcription factor(Foxp3)mRNA expression levels of each group were detected by real-time-fluorescence quantitative retroverse trascriptase polymerase chain reaction(RT-PCR).Results The primary biliary cirrhosis group showed significantly higher levels of CD4+ CD25 + Foxp3 + regula-tory T cells in peripheral blood than those of control group(P 〈0.05),and there were no difference for the mRNA level of Foxp3+ in Treg.Conclusion The expression of CD4+ CD25 + Foxp3 + T cells was decreased in patients with primary biliary cirrhosis(PBC)and weakened immune suppression function,suggesting that they may play an important role in immunopathogenesis of PBC,the level of blood Treg can refelect the cell immune state of PBC and of clinical value to&amp;nbsp;assess the prognosis.
