香菇多糖对大鼠心肌缺血再灌注损伤的保护与抗氧化作用 被引量:2

摘要 目的:探讨香菇多糖(LNT)对大鼠心肌缺血再灌注引起的氧化应激损伤的保护作用。方法:将SD大鼠随机分为5组,每组10只,分别为假手术(Sham)组,心肌缺血再灌注损伤(MIR)组,LNT低、中、高(15、30、60mg·kg-1·d-1 ig)×7d剂量组。建立心肌缺血再灌注模型。再灌注结束后采血,测定丙二醛(MDA)、超氧化物歧化酶(SOD)、天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、乳酸脱氢酶同工酶(LDHl),并测量心肌梗死面积。结果:与MIR组比较,LNT能明显降低血浆中AST、LDH、LDH1和MDA的含量,提高SOD活性,减少心肌梗死面积。结论:LNT对大鼠心肌缺血再灌注引起的氧化应激损伤有保护作用。 Objective: To investigate the protective effect of Lentinan on oxidative stress injury induced by myocardial ischemia reperfusion in rats. Methods: Fifty healthy SD rats were randomized into five groups: sham group, MIR group, MIR+high dose Lentinan treatment group, MIR+middle dose Lentinan treatment group, MIR+low dose Lentinan treatment group. Myocardial IR was carried out by ligation of left anterior descending coronary artery for 30 min followed by reperfusion for 60 min. Blood was collected at the end of reperfusion for detecting changes of aspartate aminotransferase(AST), lactate dehydrogenase(LDH), lactate dehydrogenasel(LDHI), superoxide dismutase(SOD), and malondialdehyde(MDA). The myocardial infarct size was also estimated. Results. Lentinan dose-depend-ently decreased the contents of AST, LDH, LDH1 and MDA, and the myocardial infarct size, and increased the activity of SOD. Conclusion: Lentinan exerts protective effects on myocardial ischemia reperfusion injury.
