研究生分子生物学综合实验设计──IMPACT蛋白单柱亲和纯化

摘要 设计了一个研究生分子生物学综合实验,利用内含肽介导的IMPACT系统对蛋白质进行纯化。将目的基因构建入pTWIN2载体,转化至大肠杆菌ER 2566中诱导其表达,进化亲和层析,检测其表达情况。该方法具备比以往亲和层析纯化方法更加经济的特点,可提高目的蛋白表达的成功率。通过本实验使学生掌握的分子生物学研究中最新的蛋白质分离纯化操作技术,为进一步学习和掌握复杂、综合性的分子生物学技术打下扎实的基础。 A postgraduate experiment of molecular biology was designed to express target protein in E. coli by IMPCT protein purification system. The target gene was inserted into vector pTWIN2, and then transformed to E. coli ER2566 for protein expression. This affinity purification system was a rap- id and efficient method, which could improve the solubility of target protein. By this experiment, the postgraduate students could learn the modern novel manipulation techniques of molecular biology. It would build a solid foundation for their future research.
