依达拉奉联合乌司他丁对大鼠缺血/再灌注心肌细胞凋亡的影响 被引量:2

The influence of edaravone combined with ulinastatin on ischemia/reperfusion myocardial cell apoptosis in rats

摘要 目的研究依达拉奉联合乌司他丁对大鼠缺血/再灌注心肌细胞凋亡的影响。方法将50只实验动物随机分为假手术组、缺血/再灌注组、缺血/再灌注+依达拉奉组、缺血/再灌注+乌司他丁组、缺血/再灌注+依达拉奉+乌司他丁组,采用垫扎球囊法结扎冠状动脉制作大鼠心肌缺血/再灌注模型。测定心肌组织谷胱甘肽(GSH)和丙二醛(MDA)的含量,采用TUNEL法进行细胞凋亡检测,用Western-blot方法测定凋亡因子Caspase-3、Caspase-9的表达。结果与假手术组比较,缺血/再灌注组的GSH明显减少(P<0.01),MDA及心肌凋亡指数明显升高(P<0.05),Caspase-3、Caspase-9蛋白表达增加(P<0.01);与缺血/再灌注组比较,各药物组的GSH活性显著增加(P<0.05),MDA含量及心肌凋亡指数明显减少(P<0.05),Caspase-3、Caspase-9蛋白表达降低(P<0.05),联合药物治疗组优于单独药物治疗组(P<0.01)。结论依达拉奉联合乌司他丁具有抗心肌缺血/再灌注损伤作用,其机制可能是通过调节Caspase-3和Caspase-9介导的细胞凋亡而实现。 Objective To explore the influence of edaravone combined with ulinastatin on ischemia/reperfusion myocar- . dial cell apoptosis in rats.Methods 50 cases of experimental rats were randomly divided into sham group,ischemia/ reperfusion (I/R) group,I/R+edaravon group,I/R+ulinastatin group and I/R+edaravon+ulinastatin group.The experimental model of myocardial ischemia/reperfusion in rats was established by ligating coronary artery with capsule.The content of myocardial tissue glutathione (GSH) and malondialdehyde (MDA) were determined.The cell apoptosis was detected by TUNEL and the expression of apoptosis factors Caspase-3 and Caspase-9 were determined by Western-blot.Results Compared with sham group,GSH in I/R group reduced remarkably (P〈0.0I),MDA and myocardial apoptotic index signif- icantly increased (P〈0.05),the expression of Caspase-3 and Caspase-9 enhanced (P〈0.01);Compared with the I/R group,GSH in each drug group improved (P〈0.05),MDA and myocardial apoptotic index significantly decreased (P〈0.05), the expression of Caspase-3 and Caspase-9 decreased (P〈0.05),the combined drug treatment group was superior to the single drug treatment group (P〈0.O1).Conclusion Edaravone combined with ulinastatin has the effect of anti-myocardial ischemiaJreperfusion injury.The mechanism may be related to the regulation of apoptosis mediated by Caspase-3 and Caspase-9.
