烧伤血清和痂下水肿液对内皮细胞凋亡和坏死的影响

Effects of burn serum and subeschar tissue fluid on apoptosis and necrosis of endothelial cells in vitro

目的了 解烧伤血清和痂下水肿液对内皮细胞凋亡和坏死的影响，探讨烧伤血清在烧伤后内皮细胞损伤中的作 用。方法将烧伤血清和痂下水肿液与人脐静脉内皮细胞一同孵育１２、２４ ｈ后，采用Ａｎｎｅｘｉｎ－Ｖ－Ｆｌｕｏｓ（Ａ－Ｖ）和碘化丙啶 （Ｐｒｏｐｉｄｉｕｍ ｉｏｄｉｄｅ，ＰＩ）标记内皮细胞，流式细胞仪检测其凋亡和坏死百分率。结果烧伤血清和痂下水肿液与人脐静脉 内皮细胞一同孵育 １２ ｈ后，即出现调亡和坏死增多，２４ ｈ后凋亡和坏死进一步增加。结论烧伤血清和痂下水肿液均可 导致内皮细胞凋亡和坏死，在烧伤后内皮细胞损伤中可能起重要作用。

Objective To explore the effects of burn serum and subeschar tissue fluid (STF) on postburn damage of the endothelial cells (EC). Methods Human umbilic vein ECs cultured in vitro and stained with annexin-V-Fluos (A-V) and propidium iodide (PI) were incubated with peripheral blood serum or STF collected from patients with severe burn injuries, and apoptosis and necrosis of the EC were observed by means of flow cytometry. Results Incubation with the burn serum and STF caused the increase of apoptosis and necrosis of the EC. Conclusion Burn serum and STF may induce apoptosis and necrosis of the EC, and may play an important role in postburn injury of the EC.
