摘要
目的 :探讨透明质酸酶 (HAase)和透明质酸 (HA)对血管内皮细胞增殖的影响和可能机制 ,寻找促血管新生的新途径。 方法 :采用牛主动脉内皮细胞 (BAEC)离体培养模型 ,分别经 HAase或 HA刺激 ,用 MTT法观察 BAEC增殖率的变化 ;经流式细胞术 (FCM)测定细胞周期和细胞表面粘附分子 CD44的表达。 结果 :HAase(5 0μg/ ml)作用后细胞增殖率 (% )为5 0 .10± 1.2 3(P<0 .0 1) ,使细胞周期中 S期和 G2 / M期细胞比例增加 12 .8% ,CD44的表达细胞数和表达量增加 ;而高浓度HA (10 0μg/ m l)则抑制 BAEC的增殖和 CD44的表达。 结论 :HAase可降解细胞外基质中具有抗血管新生作用的 HA,促进内皮细胞的增殖 。
Objective: To investigate the effects of hyaluronidase (HAase) and hyaluronan (HA) on proliferation of vascular endothelial cells and its mechanism. Methods: The cultured bovine aortic endothelial cells (BAEC) were treated with HAase or HA. Cell proliferation rate was detected by MTT assay. The expression of CD44 and DNA content of the cells were measured by flow cytometry (FCM). Results: HAase (50 μg/ml) stimulated cell proliferation [(50.10± 1.23)% vs control, P <0.01], increased S phase cell rate and induced the expression of CD44, but HA (100 μg/ml) inhibited cell proliferation and the expression of CD44. Conclusion: HAase may degrade antiangiogenic HA of extracellular matrix, which may stimulate proliferation of endothelial cells and enhance the curative effect of growth factors to myocardial ischemia. [
