摘要
目的研究积雪草总苷及从中分离得到的主要单体化合物对瘢痕增生的抑制作用。方法以体外培养的人增生性瘢痕成纤维细胞为试验对象,经不同浓度的样品作用后,用MTT法测定样品对瘢痕成纤维细胞增殖能力的影响,用LDH法测定样品对细胞的毒性作用。结果 8个试验药物对瘢痕成纤维细胞增殖抑制作用由强到弱的顺序为:羟基积雪草酸>积雪草酸>羟基积雪草苷>centellasaponin B>积雪草总苷>积雪草苷、积雪草苷B>centelloside C。结论羟基积雪草酸、积雪草酸、羟基积雪草苷和centellasaponin B均可抑制瘢痕成纤维细胞的增殖,且作用强于积雪草总苷。
Objective To observe the inhibition on proliferation of human hypertrophic scar fibroblasts by sapanins and triterpenoids isolated from Centella asiatica. Methods Culturing fibroblasts were treated with 8 test compound solution in different concentration. Their anti-proliferative capability was observed by MTT, and their toxicity was observed by LDH. Results The inhibition to scar fibroblast proliferation was ranged from strong to weak: madecassic acid^asiatic acid〉 madecassoside 〉 centellasaponin B 〉 total saponins 〉 asiaticoside, asiaticoside B 〉 centelloside C. Conclusion Madecassic acid, asiatic acid, madecassoside and centellasaponin B can inhibit the proliferative capability of the human hypertrophic scar fibroblasts, and are better than total saponin of Centella asiatica.
