胶原诱导关节炎大鼠CD44和Fas/FasL的表达 被引量:4

EXPRESSIONS OF CD44 AND FAS/FASL IN COLLAGEN-INDUCED ARTHRITIC RATS

摘要 目的了解胶原诱导关节炎(CIA)大鼠血清及滑膜CD44、Fas/FasL的变化及其在类风湿关节炎(RA)发病机制中的作用。方法选取雌性Wistar大鼠40只,随机分为对照组8只,CIA组32只。对照组不做任何处理,CIA组建立CIA模型,30d后测定关节炎指数(AI),用ELISA法检测血清CD44、Fas和FasL水平,RT—PCR检测滑膜CD44水平,免疫组化法检测滑膜Fas、FasL水平。结果CIA组大鼠血清CD44和Fas、FasL水平均高于对照组,差异有显著性(t=11.917~195.960,P〈0.01);滑膜CD44、Fas的表达亦明显高于对照组,而滑膜FasL的表达低于对照组,差异有显著性(t=6.916~20.264,P〈0.01)。CIA组大鼠血清CD44、Fas/FasL水平与AI呈正相关(r=0.603、0.485,P〈0.05),血清CD44水平与Fas/FasL水平呈正相关(r=0.533,P〈0.05)。结论CD44可能通过上调Fas的表达、下调FasL的表达,使得Fas和FasL结合减少,从而引起细胞凋亡减少,导致滑膜增生,参与RA的发病过程。 Objective To detect the levels of CD44 and Fas/FasL in serum and synovial tissues of collagen-induced arthritis (CIA) rat models and analyze their relation with the pathogenesis of rheumatoid arthritis (RA). Methods Forty female Wi star rats were randomly divided into two groups: control group (8 rats) and CIA group (32 rats). No any treatments were done in rats of the control group, and those in the CIA groups were created models of CIA. At 30 days after experiment, arthritis index (AD was measured, serum CD44 and Fas/FasL were detected using enzyme linked immunosorbent assay (ELISA), synovial CD44 using RT-PCR and synovial Fas/FasL using immunohistology method. Results The levels of serum CD44 and Fas/FasL in rats of CIA group were significantly higher than those of the control group, the differences were significant (t=11.917-195.960,P〈 0.01), and the levels of CD44 and Fas in the synovial tissues of CIA group were significantly higher than those of the control group, while the level of FasL was lower (t=6.916-20.264,P〈0.01). In CIA rats, a positive correlation was noted between CD44 , Fas/FasL and AI (r=0.603 ,0.485 ;P〈0.05), and between serum CD44 and Fas/FasL (r =0.533 ,P〈0.05). Conclusion CD44 is likely to be involved in the pathogenesis of RA process through raising the expression of Fas , lowering the expression of FasL, decreasing the combination of Fas and FasL, and thus reducing apoptosis , leading to synovial hyperplasia.
