摘要
目的探讨骨形态发生蛋白-7(BMP-7)对大鼠脑缺血再灌注(I-R)氧化应激损伤的影响。方法成年雄性Wistar大鼠60只,随机分为假手术组、I-R组、BMP-7组,每组20只,采用线栓法制作大脑中动脉I-R模型,观察BMP-7对大鼠神经功能缺损评分、脑梗死体积、丙二醛(MDA)含量及超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH—PX)活性的影响。结果与Ht组相比,BMP-7组大鼠神经功能缺损评分降低(t=2.33,P〈0.05),脑梗死体积减小(t=5.51,P〈0.05),MDA含量降低(F活性升高(F=22.19±68.11,q=9.41~16.25,P〈0.05)。能与清除自由基,减轻氧化性损伤有关。33.99、42.94,q=6.59、6.16,P〈0.05),SOD和GSH—PX结论BMP-7对大鼠I-R损伤具有保护作用。
Objective To investigate the protective effect of bone morphogenetic protein-7 (BMP-7) on focal cerebral ischemia reperfusion (I-R) injury in rats. Methods Sixty adult male Wistar rats were equally randomized to three groups as: sham-operated group, I-R group, and BMP-7 group. A model of I-R was created in rats by a middle cerebral artery occlusion. The effect of BMP-7 on neurological deficits, infarction volume of brain, content of malondialdehyde (MDA) , activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) were observed. Results Compared with the I-R group, neurological deficits score in BMP-7 group decreased (t = 2.33, P〈0.05), infarction volume of brain reduced (t = 5.51, P 〈0.05), the content of MDA declined (F=33.99,42.94;q=6.59,6.16;P〈0.05). the active of SOD and GPX-PX improved (F=22.19-68.11,q= 9.41-16.25,P〈0.05). Conclusion BMP-7 has a protective effect on cerebral ischemia reperfusion injury by reducing the free radicals and oxidative damage.
