氯化钆对冷保存鼠肝细胞线粒体的影响 被引量:1

摘要 目的 研究氯化钆在大鼠肝脏冷保存时对肝细胞线粒体结构、脂质过氧化物 (LPO)的影响。方法 将Wistar大鼠随机分成 8组 ,每组 5只。A、B、C、D组为对照组 ,肝脏保存时间分别为 1、2、3、4h ,其余 4组为相应的用药组 ,将各组肝脏用 0~ 4℃生理盐水灌注后于 0~ 4℃生理盐水中保存。观察各组肝细胞线粒体在电子显微镜下结构的改变 ,测定各组肝细胞线粒体脂质过氧化物含量。结果 冷保存 1、2、3h组的肝细胞线粒体脂质过氧化物的含量分别是 (1 0± 0 3)、(1 7±0 2 )、(2 0± 1 0 )nmol/g,较相应的对照组的含量显著降低 (2 2± 1 0、2 8± 1 0、3 5± 1 0 ,P <0 0 5 )。随着冷保存时间的延长 ,对照组肝细胞线粒体损伤逐渐加重 ,表现为肝细胞线粒体肿胀 ,内膜嵴断裂 ,空泡形成 ,严重者线粒体结构基本被破坏。氯化钆明显减轻肝细胞线粒体结构的损伤程度。结论 线粒体损伤是大鼠肝脏冷保存的特征 ,氯化钆对冷保存鼠肝肝细胞的脂质过氧化损伤有抑制作用。 Objective[WT5”BZ] To study the effect of gadolinium chloride hexahydrate (GDCL 3) on the structure and lipid peroxide (LPO) of rat hepatocellular mitochondria during liver cold ischemia storage. [WT5”HZ] Methods[WT5”BZ] Forty Wistar rats were randomized into 8 groups ( 5 in each group). Group 1~4 served as control. The other 4 groups were pretreated with GDCL 3 (7?mg/kg). The ultrastructures of hepatocellular mitochondria were observed by electron microscope and LPO of hepatocellular mitochondria were investigated. [WT5”HZ] Results[WT5”BZ] LPO of hepatocellular mitochondria in group 1?h, 2?h, 3?h pretreated with GDCL 3 for 1、 2、 3 h were 1 0±0 3、1 7±0 2、2 0±1 0, LPO was suppressed significantly compared with control group (2 2±1 0、2 8±1 0、3 5±1 0、 P <0 05). With time prclonging of liver storage, the damage of hepatocellular mitochondria ultrastructure became more severe and LPO of hepatocellular mitochondria increased significantly. Pretreated with GDCL 3, the damage of hepatocellular mitochondria ultrastructures were lessened and the elevation of LPO was suppressed significantly (1 hour、 2 hours、3 hours, P <0 05). [WT5”HZ] Conclusion[WT5”BZ] Mitochondria damage is one of the sensitive characters of liver cold ischemia injury, GDCL 3 inhibits rat hepatocellular injury during liver cold ischemia storage.
