榄香烯对HeLa细胞端粒酶催化亚单位基因表达的作用 被引量:14

摘要 目的 :探讨HeLa细胞凋亡过程中 ,人乳头瘤病毒 18型E6 (HPV18 E6 )基因、p5 3基因、端粒酶催化亚单位 (hTERT)基因表达的变化 ,为临床诊断、治疗宫颈癌提供新思路。方法 :用流式细胞仪、电镜证实细胞凋亡 ;用PCR和RT PCR方法研究HPV18 E6基因、p5 3基因、hTERT基因表达在细胞凋亡过程中的变化及相互关系。结果 :在榄香烯作用下 ,HPV18 E6基因、p5 3基因表达没有变化 ,但hTERT基因表达受到抑制。结论 :榄香烯诱导HeLa细胞凋亡过程中 ,hTERT基因表达受到明显抑制 ,对于耐药性肿瘤 。 Purpose:The experimental study on the changes of HPV18 E6 gene,p53 mRNA and telomerase catalytic subunit (hTERT) mRNA in the process of HeLa cell apoptosis was carried out to supply a new way to diagnosing and treating cervical cancer. Methods:The apoptosis of HeLa cell was confirmed by means of electron microscope and flow cytometry. With PCR and RT PCR, we studied the changes and relations among HPV18 E6 DNA,p53 mRNA as well as hTERT mRNA in the process of HeLa cell apoptosis. Results:HPV18 E6 DNA and p53 mRNA had no changes but hTERT mRNA activation was inhibited by elemene emulsion. Conclusions:During apoptosis of HeLa cells induced by elemene emulsion, hTERT mRNA activation could apparently be inhibited. Hence, inhibition of telomerase activation may represent a new chemosensitisation for tumors resistant to anticancer drugs.
